TOFA Compound Boosts Fat Burning and Preserves Muscle in Mice

What the Study Found About TOFA

A research team at the University of California, Berkeley tested the compound 5-tetradecyloxy-2-furoic acid (TOFA) in obese mice. They found that TOFA increased energy expenditure by up to 18%, reduced body fat, and did not cause significant muscle loss.

TOFA also improved blood sugar levels, lowered triglycerides, and reduced signs of fatty liver disease in the mice. The compound worked by activating genes that promote fat burning and energy production.

How TOFA Works Differently From Current Obesity Drugs

Current GLP-1 medications like Ozempic, Wegovy, Mounjaro, and Zepbound help people lose weight mainly by reducing appetite. However, these drugs can cause side effects such as nausea and may lead to loss of muscle mass.

TOFA takes a different approach by increasing the amount of energy the body uses, rather than simply reducing food intake. It acts as an ACC inhibitor and also activates PPARα and PPARδ receptors, which are involved in fat metabolism.

Evidence and Study Details

The findings come from preclinical experiments in mice, not humans. The study was published in Science Advances on August 21, 2026.

Researchers compared TOFA to other ACC inhibitors and found that, unlike some similar compounds, TOFA did not raise triglyceride levels. They also tested combining TOFA with GLP-1 drugs and found the combination produced stronger results in mice.

Implications for Metabolic Health Management

For people managing obesity, diabetes, or fatty liver disease, a treatment that burns fat without causing muscle loss could address some limitations of current therapies.

The study suggests that targeting both energy expenditure and fat metabolism may be more effective than focusing on appetite alone. However, these results are limited to animal models so far.

Limitations and Next Steps

TOFA has not been tested in humans, and its safety and effectiveness in people are unknown. Previous ACC inhibitors have not been approved for metabolic disease due to concerns about side effects.

The researchers note that more studies are needed to determine if TOFA’s benefits in mice will translate to human treatments. Until then, TOFA remains an experimental compound.
