Potential Side Effects of Unapproved GLP-1 Drug
University of Pennsylvania researchers used machine learning to track self-reported reactions to gray-market retatrutide on Reddit.
AI Analysis Evaluates Potential Side Effects of Unapproved GLP-1 Drug
Researchers at the University of Pennsylvania used artificial intelligence (AI) to systematically review online forum discussions about the potential side effects of unapproved GLP-1 drug formulations. The investigators analyzed nearly 150,000 posts and comments from roughly 40,000 Reddit users to identify self-reported health experiences with retatrutide.
Retatrutide is an investigational glucagon-like peptide-1 (GLP-1) medication that is not approved for human use by regulatory agencies. While pharmaceutical clinical trials remain underway, some individuals purchase gray-market chemical versions not intended for human consumption and share their reactions online.
Reported Symptoms Differ from Phase 2 Clinical Trial Records
Increased appetite was the most frequently self-reported symptom among online forum participants who described taking unapproved retatrutide. This pattern contrasts with formal Phase 2 clinical trial findings, where gastrointestinal complaints represented the most common side effects.
The computational research team identified approximately 14,000 users who claimed personal use of the unapproved compound. Using standardized medical terminology, the researchers isolated 7,823 users who reported at least one classifiable symptom beyond expected changes such as weight loss or appetite suppression.
Gray-Market Variables Cloud Potential Side Effects of Unapproved GLP-1 Drug
Online self-reports cannot confirm whether the reported symptoms stem from retatrutide or from product impurities. Co-author Lyle Ungar, a professor in Computer and Information Science (CIS) at the University of Pennsylvania, emphasized that forum participants may not be taking genuine retatrutide because gray-market product formulations vary widely.
The research team noted that social media observations cannot establish drug safety, symptom causation, or clinical equivalence with trial participants. Lead author Neil Sehgal stated that the study in JAMA Network Open provides an initial observational glimpse into an unmonitored population rather than definitive clinical proof.
Mechanisms and Clinical Research Next Steps
Retatrutide targets three separate hormone receptors that regulate metabolic function, appetite, and blood glucose, whereas semaglutide targets one and tirzepatide targets two. The multi-receptor mechanism prompted researchers to test whether user experiences would diverge from approved single-target or dual-target treatments.
The research group previously used large language models (LLMs) across 400,000 posts regarding semaglutide and tirzepatide to detect underreported issues like menstrual irregularities. Public health scientists can use these computational findings as exploratory hypotheses for future clinical monitoring. Anyone evaluating novel weight-loss compounds should speak with a licensed clinician about the documented risks and potential side effects of unapproved GLP-1 drug products.
What to Look For in a Body-Composition Scale
A bioimpedance scale trends body fat and muscle rather than just weight — useful when the number stalls but your composition is improving. The absolute body-fat % isn't lab-accurate, so use it for TRENDS at a consistent time of day. Choose one that syncs to an app so you can watch the trend line.
