Study Finds Semaglutide Extends Lifespan in Mice
A study published in Nature reported that semaglutide, a glucagon-like peptide-1 (GLP-1) drug, extended the median lifespan of older, healthy female mice by nearly 100 days compared to untreated controls.
Researchers at the University of California, Berkeley, administered semaglutide to 20-month-old female mice for three months, then followed some animals until natural death to assess lifespan effects.
GLP-1 Treatment Improved Muscle, Cognition, and Aging Markers
Mice treated with semaglutide showed improvements in muscle function and cognitive performance compared to untreated mice.
Gene expression analysis revealed that semaglutide reduced hallmarks of aging, such as increased inflammation and decreased regenerative capacity.
Comparison With Calorie Restriction Shows Unique Effects
Researchers compared semaglutide treatment to a 24% calorie-restricted diet in similarly aged mice. Both groups showed stable physiological measurements, but semaglutide-treated mice had greater improvements in exploratory behavior, spatial memory, and blood sugar regulation.
Unlike calorie restriction, semaglutide did not lower metabolic rate, suggesting the drug may act through a different biological pathway than dietary interventions.
Implications and Limitations for Human Metabolic Health
The study suggests that GLP-1 drugs like semaglutide may slow physiological aging in mice, potentially explaining their broad benefits in delaying age-related diseases in animal models.
However, the authors and outside experts caution that these findings cannot be directly applied to humans. Clinical trials are needed to determine if GLP-1 drugs have similar effects on human lifespan or healthy aging.

GLP-1 drugs are already used in humans for blood sugar control and weight management, but their use for aging or lifespan extension is not approved or established.
