What a 99% Peptide Purity Test Result Leaves Out

Most certificates report a single number, and five separate assays sit behind the question it is asked to answer.
A peptide purity percentage compares the target peptide against other peptide-related material. It says nothing about identity, dose, sterility or how much of the powder is salt and water.
A 99% purity result tells you what percentage of the peptide-related material in a vial is the peptide you ordered. We regularly read these certificates for the peptide guides we publish here, and this number is consistently given weight it was never meant to carry.
A certificate that shows a high purity figure, and nothing else, answers just one question out of five. Freedom Diagnostics says it never issues a purity result without accompanying mass spectrometry identity confirmation. Most vendor certificates omit that pairing.

What the Purity Percentage Measures
Purity on a peptide COA is a chromatography result, usually from high-performance liquid chromatography (HPLC). The sample is pushed through a column that separates molecules by how strongly they stick to it, and a detector records a peak for each one. Purity is the area of the target peak divided by the total area of all peaks, expressed as a percentage.
The impurities that show up are the ones synthesis leaves behind. Chains that stopped a residue short, chains that never had a protecting group removed, and fragments that broke during handling all appear as their own peaks. A 95% result means those by-products account for one twentieth of the peptide material present.
For most research peptides the usable range starts around 95%. Reputable synthesis targets 98% or 99%. The difference between 95% and 99% matters less than most buyers assume, because the impurities are chemically similar fragments of the same molecule. Act on a result under 90%, which suggests synthesis or storage went wrong.
Chromatographic Purity Is Not Peptide Content
The purity percentage ignores everything in the vial that is not a peptide. Product pages rarely mention it, and it changes the arithmetic on every dose you draw.
Lyophilized peptide powder carries bound water, residual solvents and salt counterions, most often trifluoroacetate (TFA) left over from purification. None of that registers on the HPLC trace, so none of it reduces the purity figure. The separate measurement that captures it is called net peptide content, and as AmbioPharm sets out, typical values run between 70 and 90 percent.
Work that through and a 10 mg vial at 99% purity and 80% peptide content holds roughly 8 mg of peptide. Peptides rich in arginine bind more counterion and sit at the low end of that band. Calculate a milligram dose from vial weight alone and you under-dose by a fifth. The certificate that said 99% never disagreed with you.
Vanguard Laboratory sells this as a separate line item. It describes its mid-tier service as covering identity, purity and net peptide content. If a COA reports only purity, the content number was not measured.
Identity, Potency and Sterility Are Separate Assays
Four more questions sit outside the purity number, and each needs its own test:
- Identity confirms the peak is the molecule named on the label. Mass spectrometry weighs the molecule and matches it to the expected mass. Without it, a 99% pure something is still a 99% pure something else.
- Quantity confirms the vial holds the milligrams the label claims. Underfilling is a documented failure in this market and purity cannot detect it, because a half-full vial of pure peptide is still pure.
- Sterility and bioburden confirm nothing is growing in the powder. This one matters most, since these products are injected and skip the defenses that stop a swallowed contaminant.
- Endotoxin measures bacterial cell wall fragments, which survive sterilization and cause fever and inflammatory reactions even when nothing viable is left.
Heavy metals and residual solvents are two further screens, usually sold separately again. A vendor certificate showing one number has skipped the assays that catch sterility and dose failures.
How to Read a Certificate of Analysis
Work down a COA in the same order every time, and most bad ones fail within the first three checks:
- The laboratory name. A certificate issued by the seller's own quality department is a self-report. Look for an independent laboratory on the letterhead.
- The lot or batch number. It must match the number printed on the vial you were sent. A certificate for a different lot describes a different powder.
- The date. Peptides degrade, and a result from two years ago describes the batch on the day it was made.
- The methods listed. HPLC alone gives purity. Look for a mass spectrometry line for identity, and named methods for anything else claimed.
- The peaks reported. A real chromatogram shows the trace itself. A single unexplained number with no underlying data is a marketing asset.
- Verification. Freedom Diagnostics publishes every COA it issues in a public lookup that takes an accession number or a company name. Janoshik reports can be checked at its verification page using the code printed on them. If a certificate cannot be checked at the issuing lab, treat it as an image file.
Forged and recycled certificates circulate in this market, so verify the document at the laboratory that issued it. A PDF is trivial to edit. A record in the issuing laboratory's own database is not.
What a Certificate Cannot Tell You
A COA describes one sample from one batch on one day, and your vial is not that sample. Even a genuine, current, independently issued certificate leaves the last step unverified, because whoever filled your vial could have drawn from a different container.
Storage sits outside it too. A peptide synthesized and tested correctly, then shipped through a warm sorting facility or left at room temperature for a fortnight, is no longer the substance the certificate described. Nothing on the document covers what happened after it was signed.
The strongest version of the check is therefore testing the vial you were sent, rather than reading the certificate the seller supplied. It costs more and it takes days. It is also the only version that describes your peptide.
How to Get One Vial Tested
Consumer peptide testing is a real service with published prices, and you do not need to be a laboratory to use it. Vanguard Laboratory in Olympia, Washington lists purity alone at $200 and purity with quantity at $250. Endotoxins are $210, sterility is $150 and heavy metals are $150. Turnaround at its entry service runs 5 to 8 days. Freedom Diagnostics in Franklin, Tennessee states an average of 2 to 4 days.
The process is the same everywhere: order the test, ship the sealed vial with the labeling the lab specifies, and receive a digital report. Most labs want the vial unopened, and several require more than one vial for the multi-assay packages, so buy with that in mind if you intend to test.
There is also a free route. Finnrick tests consumer samples at no charge for US residents and publishes the results as vendor rankings, with 9,993 tests recorded as of 28 August 2026. Who Should Not Pay for Independent Testing
If you are getting peptides on prescription through a compounding pharmacy, paying a second lab is usually redundant. The pharmacy already owes you a certificate for the lot it dispensed, and a 503B outsourcing facility operates under good manufacturing practice with batch records behind it. Ask for the document first, and only pay for a test if the answer is no.
The arithmetic also fails on small orders. A $250 purity and quantity test on a $60 vial costs four times the product. At that ratio the money goes further on a prescription route, where the testing is someone else's obligation. Testing earns its cost when you are evaluating a source you intend to keep using. On a single vial it rarely does.
Nobody should read a clean COA as clearance to inject something. A certificate speaks to what is in the powder. Whether the compound is appropriate for you is a separate question, and it belongs with a clinician who knows your history.
What Would Change This Answer
A required assay panel would change it. Suppose sellers had to publish identity, quantity, sterility and endotoxin results per lot rather than one purity figure. Reading a certificate would stop being a skill, and the gap would close.
Wider legal compounding access would also change it. Move the compounds people currently source as research chemicals onto a lawful prescription route, and the testing obligation shifts to a licensed pharmacy. Until then, treat peptide purity testing as four questions rather than one. Pull the COA for your specific lot and check that the issuing laboratory can verify it. Then take the numbers to the clinician supervising your protocol, before you draw a dose.
